The obtention of an appropiate substrate is a major concern for scientists in the investigation of enzymatic activity. Beside the basic features required for HTS applications (sensitivity, robustness and speed) an increasing interest is also shown nowadays for high specific substrates with a structure as close as possible to the natural ones. Moreover substrates structure gives very valuable information for the research of inhibitors.
In this respect, PHARMALEADS proposes:
The detection, thanks to its original technology and a library of 5000 substrates, of an appropriate substrate for any given/known protease, with required features such as sensitivity, robustness, good kinetic parameters to perform H.T.S. (FLUOFAST)
The optimization of any given substrate for proteases i.e. modification of the structure of an existing substrate to make it closer to the natural one which will improve its specificity and (or) kinetic parameters.
The design of potent inhibitors in the field of metallopeptidases.
The access to a catalogue of specific substrates for metallopeptidases as ECE, NEP, ACE, Botulinum Toxins A and B as well as for the phosphatases PTP1B and CD45.
